We have a customer where we have installed a new Primary Site yesterday with SCCM SP2 integrated version. After starting to configure it, I saw that Multicast and Virtual Apps options are not in place.

I assume that these options are in R2 only, correct? My question is now, can I install R2 over SP2 or is this not supported?

I didn’t find a usable link via Google which says this is possible.

First things first yes Multicast and Virtual Apps are R2 features hence why you are not seeing them.  Second yes you can install R2 post SP2 after which point you should see these options.
